2018年2月23日 · In this study we employ magnetic force microscopy (MFM), transmission electron microscopy (TEM), and electron energy loss spectroscopy (EELS) to characterize differences in ferritin (iron) distribution and composition across injured and non-injured tissues by employing a rodent model of spinal cord injury (SCI).

To our knowledge, this is the first study to provide a direct comparison of three frequently used histochemical iron stains on human paraffin-embedded AD brain tissue and simultaneously investigate changes in the distribution of ferritin. We evaluated their ability to stain iron-containing plaques and microglia as well as myelin-associated iron.

We show that ceruloplasmin (Cp), a ferroxidase that oxidizes toxic ferrous iron, is important for this process. SCI in Cp-deficient mice demonstrates that Cp detoxifies and mobilizes iron and reduces secondary tissue degeneration and functional loss. Our results provide new insights into how astrocytes and macrophages handle iron after SCI.

Superficial siderosis of the CNS is an unusual disorder in which iron is deposited around involved structures in the brain, spinal cord, and some cranial nerves. Clinically, superficial siderosis manifests as a triad of hearing loss/tinnitus, cerebellar ataxia, and myelopathy, typically years after the initial bleed.

Excess iron accumulation within the spinal cord is thought to exacerbate tissue damage and limit functional recovery after traumatic spinal cord injury (SCI). An optimal treatment to reverse or prevent damage would be to deliver an iron chelator systemically.

Histologic sections of the excised cords, which were stained for myelin, iron, and cell bodies (Nissl), were used for correlation to the images. We found that significant Fourier truncation and partial-volume imaging artifacts modulated the MR display of the cord.
